Kurzfassung
Due to the reduced ability of hyaline cartilage to heal itself, cartilage defects should be treated to prevent the onset of osteoarthritis. The aim of this study is to compare two novel BG scaffolds with a patented ion composition tailored for cartilage with no (CAR12N) or low calcium oxide/magnesium oxide content (CAR12N + CaO/ MgO) with the conventional BG1393 with the same topology. The scaffolds consisting of 100% BG are characterized and dynamically seeded with primary porcine articular chondrocytes (pAC) or primary human mesenchymal stem cells (hMSC). Vitality, metabolic activity, DNA, sulfated glycosaminoglycan (sGAG), cartilagespecific gene and protein measurements are performed. Porosity of all scaffolds is over 65%. pH profiles confirm ion release. The vitality of pAC colonized CAR12N is over 80% after 7 and 21 days. Metabolic activity is maintained by both cell types on all scaffold variants. The sGAG amount per pAC is significantly higher in BG1393 compared to CAR12N and CAR12N + CaO/MgO. Cartilage-specific genes are expressed in all pAC or hMSC colonized scaffold variants. Cartilage-specific matrix components are detected at protein level in pAC and hMSC colonized CAR12N scaffolds. In conclusion, CAR12N maintains the chondrogenic phenotype of pAC or supports features of chondrogenesis of hMSC and thus, shows high potential for cartilage repair.
