The study finds that the ZBTB16/CUL3/ROC1 ubiquitin ligase complex directly drives the accelerated degradation of pathogenic pendrin (SLC26A4) protein variants, revealing the precise molecular mechanism underlying protein loss-of-function in Pendred syndrome and genetic hearing loss. By seamlessly combining diverse experimental biology techniques with advanced AI-based structural modeling, this interdisciplinary collaboration successfully maps complex molecular interactions to pinpoint novel targets for therapeutic intervention.
